With classic floors like this you need to be a little careful because most will be fixed with bitumen adhesive which is also the Damp Proof Membrane.
Without knowing the cause of the stain on the floor (water, oil, bleach etc etc) and the type of wood you should not proceed with DIY remedial actions as this could cause further problems. Although I am biased here I would suggest a professional floor sander/sealer has a look. They may be able to offer more precise advice on how to proceed. Look for someone who specialises in wood not a generic flooring fitter.
